Setting Tiny Core passwords

I read on the wiki how to activate passwords. But my question is: Where is it better to activate them?
1) In Extlinux?
2) In TinyCore as root?
3) In TinyCore as user tc?
4) The two previous ones?
Basically I would like have one password to enter the computer and another one as superuser (f.i. sudo su <password>), but I have the doubt if root = superuser or they are different users.

Yes, root and superuser are the same user. That users home directory is:

Some programs will store settings there if run as root. Midnight commander (mc) is one such program. If you want those settings
remembered the next time your machine reboots, add root to your /opt/.filetool.lst file so it gets backed up.
